Don't know about internal difference,and don't think there are any. The main difference is the frame's grip angle. The 22/45 is made to be just like a 1911 pistol and they do have a model that 1911 grip panels fit. I think the grip on the MkIII is just like the MkI and II.
Which is better, depends on which one you like best. :)

NRA_guy brought up the configurations. With the MkIII you have more to consider. I wish my 28 year old MkII had the Hi-Viz front sight but I haven't changed it yet. I'm still considering a new MkIII though to continue series. Need my MkII Standard back.

also the 22/45 is a polymer frame, not metal like the mkIII... but a bull barreled target 22/45 is ~$100 cheaper than the same model in the mkIII. There was a mkII 22/45 first, but the current is based on the mkIII's. pretty much all internals are the same except for the bolt release and the magazines
